There are 14 copies of ISKpn74 in the chromosome of K. pneumoniae PBIO2030. Two adjacent insertion sequences caused an inversion, backed up by the shuffling of direct repeats. The direct repeat in the left flank of the first insertion sequence (AGGAACGGC) is identical to the direct repeat in the right flank of the second insertion sequence and vice versa (TATTTTTGG). Additionally, K. pneumoniae PBIO2030 harbors a chromosomally inserted virulence plasmid which is framed by ISKpn74. The direct repeat in the right flank of one insertion sequence is identical to the direct repeat in the left flank of another insertion sequence (ATTTTCGCG). This is explainable by a co-integrate formation involving both elements. Please observe that the other two flanks share no identical direct repeat, possibly because of a deletion event involving yet another insertion sequence. Two IS elements which are located in the inserted plasmid share the same SNV (single-nucleotide variant; G->T, Lys3Asn) at position 86.
